目的观察加味镇眩颗粒对自发性高血压大鼠(SHR)血浆血管紧张素Ⅱ(AngⅡ)、心肌基质金属蛋白酶-2(MMP-2)表达水平和微血管密度(MVD)的影响。方法将50只SHR大鼠随机分为加味镇眩颗粒高剂量组、加味镇眩颗粒中剂量组、加味镇眩颗粒低剂量组、卡托普利组、SHR对照组,经过12周的干预,分别测定动物血压变化,血浆AngⅡ含量、免疫组织化学法检测心肌MMP-2蛋白表达水平、心肌微血管密度。结果治疗组均可降低SHR血压、血浆AngⅡ含量、心肌MMP-2蛋白表达水平,增加SHR心肌微血管密度,与SHR组相比差异具有统计学意义(P<0.05),中药组中,加味镇眩颗粒高剂量组效果最好。与卡托普利组相比,高剂量组在12周末降压效果相当(P>0.05),且更能降低心肌MMP-2蛋白表达水平(P<0.05),增加SHR心肌微血管密度(P<0.05)。结论加味镇眩颗粒可降低SHR血压,可能与降低血浆血管紧张素Ⅱ含量,下调心肌MMP-2蛋白表达水平,改善SHR大鼠心肌微血管稀疏的作用有关。
Objective To observe the effects of Jiawei Zhenxuan Granule on the plasma levels of angiotensin Ⅱ (Ang Ⅱ), matrix metalloproteinase-2 (MMP-2) and microvessel density (MVD) in spontaneously hypertensive rats (SHR) Methods A total of 50 SHR rats were randomly divided into four groups: Jiawei Zhenxuan Granule high dose group, Jiawei Zhenxuan Granule middle dose group, Jiawei Zhenxuan Granule low dose group, Captopril group and SHR control group. After 12 weeks of intervention, The changes of blood pressure and plasma AngⅡ were measured respectively. The expression of MMP-2 protein and myocardial microvessel density were detected by immunohistochemistry. Results In the treatment group, SHR blood pressure, plasma AngⅡ content, myocardial MMP-2 protein expression and myocardial microvessel density were increased in SHR group compared with those in SHR group (P <0.05) High-dose group of particles the best effect. Compared with captopril group, the high-dose group had a similar antihypertensive effect on the 12th week (P> 0.05), moreover, it decreased myocardial MMP-2 protein expression (P <0.05) and increased myocardial microvessel density (P < 0.05). Conclusion Jiawei Zhenxuan Granule can reduce the blood pressure of SHR, which may be related to the decrease of plasma angiotensin Ⅱ, the down-regulation of MMP-2 protein expression and the improvement of myocardial microvascular thinning in SHR rats.
