目的:探讨抑癌基因Rb、p16和癌基因CyclinD1在大肠癌中的表达及临床意义。方法:采用流式细胞分析技术(Flowcytometry,FCM),定量研究在大肠癌与正常粘膜中Rb、p16、CyclinD1的表达情况,所有数据换算成荧光指数(FluorescenceIndex,FI)值后,利用SAS软件包进行统计分析。结果:1)30例大肠癌组织与正常粘膜中Rb、p16、CyclinD1FI(x±s)值经t检验,均P<0.01;2)以Rb表达阳性、阴性进行分组,在Rb阳性下,p16与组织学分级呈负相关,CyclinD1与组织学分级呈正相关,p16与淋巴转移呈负相关,均P<0.01。在Rb阴性下,Rb与淋巴转移呈负相关,p16与淋巴转移呈正相关,而CyclinD1与淋巴转移呈负相关,均P<0.01。结论:1)Rb、p16、CyclinD1在大肠癌、正常粘膜组织中的表达存在显著差异;2)在Rb阳性下,p16失活、缺失或突变会导致组织的恶性程度增加且易发生淋巴转移,CyclinD1的激活会导致组织的恶性程度增加;3)在Rb阴性下,各种结果显示:负反馈机制在其中可能起作用。
Objective: To investigate the expression of tumor suppressor gene Rb, p16 and oncogene CyclinD1 in colorectal carcinoma and its clinical significance. Methods: Flowcytometry (FCM) was used to quantitatively study the expression of Rb, p16 and CyclinD1 in colorectal carcinoma and normal mucosa. All the data were converted into FluorescenceIndex (FI) values and analyzed by SAS software package conduct statistical analysis. Results: 1) The expression of Rb, p16 and CyclinD1FI (x ± s) in 30 cases of colorectal cancer and normal mucosa were all significantly lower than those in normal mucosa (all P <0.01); 2) Rb was positive and negative, There was a negative correlation between histological grade and histological grade. CyclinD1 was positively correlated with histological grade, while p16 was negatively correlated with lymphatic metastasis (all P <0.01). In Rb negative, Rb was negatively correlated with lymphatic metastasis, p16 was positively correlated with lymphatic metastasis, while CyclinD1 was negatively correlated with lymphatic metastasis, both P <0.01. Conclusions: 1) The expression of Rb, p16 and CyclinD1 in colorectal cancer and normal mucosa is significantly different; 2) In Rb positive, p16 inactivation, deletion or mutation will lead to the increase of malignancy and lymphatic metastasis, Activation of CyclinD1 leads to an increase in tissue malignancy; 3) Under Rb-negative, various results show that a negative feedback mechanism may play a role in this.
