目的:观察半夏泻心汤对伊立替康(irinotecan,CPT-11)致小细胞肺癌(small-cell lung cancer,SCLC)荷瘤鼠胃损伤是否具有改善作用。方法:建立SCLC荷瘤鼠,将40只荷瘤鼠随机分成4组,高、中、低3个给药组半夏泻心汤剂量分别为20、10、5g.kg-1.d-1,空白组给等量水,灌胃给药,共8天。第3天开始尾静脉注射CPT–11,35 mg.kg-1.d-1,共4天。观察荷瘤鼠食量情况;第8天处死荷瘤鼠,取胃组织,显微镜下观察其病理形态改变,免疫组化测胃黏膜环氧化酶-2(Cyclooxygenase-2,COX-2)表达;另取胃组织,行组织匀浆,离心,酶联免疫吸附法(Enzyme-linked immunosorbent assay,EL ISA)检测胃组织中血栓烷B2(TXB2)和6-酮-前列腺素F1α(6-keta-PGF1α)含量;取舌黏膜组织,显微镜下观察其病理形态改变。结果:半夏泻心汤给药组相较于空白组食量增加,空白组胃黏膜内及黏膜下少量血管有扩张,稍有水肿表现,舌黏膜空白组表皮完整,少量细胞空泡变性,鳞状上皮增生,表面角质层增厚,给药组均好于空白组。胃组织中TXB2和6-keta-PGF1α含量及胃黏膜COX-2表达情况各组无统计学差异。结论:半夏泻心汤对CPT-11致SCLC荷瘤鼠胃损伤具有一定的改善作用。
Objective: To observe whether Banxiaxiexin Decoction can improve the gastric injury of small cell lung cancer (SCLC) induced by irinotecan (CPT-11). Methods: SCLC tumor-bearing mice were established. Forty tumor-bearing mice were randomly divided into 4 groups. The dosage of Banxiaxiexin Decoction of high, middle and low dosage groups was 20, 10, 5g.kg-1.d-1 , Blank group to the same amount of water, gavage, a total of 8 days. On the third day, tail vein injection of CPT-11,35 mg.kg-1.d-1 was started for 4 days. The tumor-bearing mice were sacrificed on the 8th day. The tumor-bearing mice were sacrificed and the gastric tissues were taken out. The pathological changes of the tumor-bearing mice were observed under a microscope. Results: Compared with the control group, the food intake of Banxiaxiexin Decoction group was increased, the number of blood vessels in the gastric mucosa and submucosa of the blank group was dilated and slightly edematous, the epidermis of the tongue mucosa blank group was intact, the number of cells vacuolar degeneration, Epithelial hyperplasia, the surface of the stratum corneum thickening, the administration group were better than the blank group. The content of TXB2 and 6-keta-PGF1α in gastric tissue and the expression of COX-2 in gastric mucosa had no statistical difference. Conclusion: Banxiaxiexin Decoction has a certain effect on gastric injury induced by CPT-11 in mice with SCLC.
