目的研究建立免疫亲和柱净化柱前衍生-高效液相色谱-荧光检测法,测定坚果类食品中的黄曲霉毒素B1、B2、G1、G2的快速而准确的方法。方法样品经过甲醇-水(7+3)提取后,通过免疫亲和柱净化,柱前衍生,高效液相色谱-荧光检测器测定。结果在优化条件下,黄曲霉毒素B1、B2、G1、G2的检出限分别为0.2、0.05、0.2、0.05μg/kg,有较好的准确度和精密度,回收率为82%~92%,相对标准偏差<5%。结论该法操作简便、灵敏、准确、无杂质干扰,适合坚果类食品中黄曲霉毒素的日常检测。
Objective To establish a rapid and accurate method for the determination of aflatoxins B1, B2, G1 and G2 in nut foods by using immunoaffinity column to purify pre-column derivatization-high performance liquid chromatography-fluorescence detection. Methods The samples were extracted with methanol-water (7 + 3) and purified by immunoaffinity column, pre-column derivatization and high performance liquid chromatography-fluorescence detector. Results Under the optimum conditions, the detection limits of aflatoxins B1, B2, G1 and G2 were 0.2,0.05,0.2,0.05μg / kg respectively, with good accuracy and precision, and the recoveries were 82% ~ 92 %, Relative standard deviation <5%. Conclusion The method is simple, sensitive, accurate, no impurity interference, suitable for routine testing of aflatoxins in nuts.
