目的 :观察 P-糖蛋白基因疫苗免疫小鼠诱导的免疫病理变化。方法 :P-糖蛋白基因疫苗 2 0~ 1 0 0 μg两次肌注 (间隔 9d)免疫小鼠 ,收集血清检测特异性抗体效价 ;取心、肝、肾制作组织切片 ,HE染色行病理检查。结果 :末次免疫后 5 d鼠血清特异性抗体效价达到 1∶ 4 0 0 0 ;心、肾组织正常 ,但肝脏出现自身免疫性肝炎改变。结论 :P-糖蛋白基因疫苗免疫小鼠可诱导实验性肝炎 ,实验性肝炎小鼠可作为人自身免疫性肝炎的动物模型
Objective: To observe the immunopathological changes induced by P-glycoprotein gene vaccine in mice. Methods: The mice were immunized with P-glycoprotein gene vaccine 20 ~ 100 μg twice intramuscularly (interval 9 days). The serum was collected to detect the titer of specific antibody. Tissue sections of heart, liver and kidney were made and stained with hematoxylin and eosin an examination. Results: The titer of serum specific antibody reached 1: 40000 at the 5th day after the last immunization. The heart and kidney tissues were normal, but the autoimmune hepatitis in the liver was changed. CONCLUSION: Experimental hepatitis can be induced in mice immunized with gene P-glycoprotein and experimental mice can be used as animal models of human autoimmune hepatitis
