目的:观察ETB受体激活对猪冠状动脉阻力血管有何影响。方法:采用电视显微镜观察离体猪冠状动脉阻力血管内径。实验分为3组。各组分别在对照观察、内皮破坏或105mol/LL-N-硝基精氨酸甲酯(L-N-nitro-arginingmethyLester,L-NAME)处理后,给予内皮素B受体(endothelin-Breceptor,ETB)激活剂sarafotoxin6c(SF6c),记录浓度-反应曲线。结果:给予SF6c后,对照组血管呈现浓度依赖性轻度舒张及轻度收缩反应:内皮破坏组及一氧化氮合成酶抑制组仅表现为浓度依赖性收缩反应。结论:ETB受体激活可对冠状动脉阻力血管产生收缩舒张双重作用;其中舒张作用依赖内皮存在和一氧化氮的产生
Objective: To observe the effect of ETB receptor activation on the vascular resistance of porcine coronary artery. Methods: Observe the vascular diameter of isolated porcine coronary artery with TV microscope. The experiment is divided into 3 groups. Each group was given endothelin-B receptor (L-NAME) after the control observation, endothelium destruction or L-N-nitro-arginine methylester (L-NAME) Breceptor, ETB) activator sarafotoxin6c (SF6c), record the concentration-response curve. Results: After administration of SF6c, the blood vessels in the control group showed a concentration-dependent mild relaxation and mild contractile responses: the endothelial dysfunction group and the nitric oxide synthase inhibitor group showed only a concentration-dependent contraction response. CONCLUSION: ETB receptor activation may play a dual role in the contraction and relaxation of coronary resistance vessels. The relaxation effect depends on the presence of endothelial cells and the production of nitric oxide
