为探讨人载脂蛋白B基因3’端高可变区(hypervariableregion,HVR)不同等位基因对真核细胞基因表达调控的作用.选取在正常人群中分本最广的HVE36小等位基因和在冠心病病人中分布占优势的大等位基因HVE44,分别克隆进含荧光素酶基因的pGL2-promoter及pGL2-control质粒中,转染Hela细胞和HepG2细胞.结果发现,在不同的细胞系中插入HVE44等位基因的重组质位表达均高于插入HVE36的重组质粒的表达。表明载脂蛋白B基因3’端高可变区不同等位基因具有调控基因表达的作用,HVE44大等位基因可上调细胞内的基因表达,这或许是以HVE44等大等位基因为主体的动脉粥样硬化患者体内载脂蛋白B升高的原因之一.
To investigate the effect of different alleles of the 3 ’hypervariable region (HVR) of human apolipoprotein B gene on the regulation of gene expression in eukaryotic cells. Select the most widely distributed HVE36 allele in normal population and HVE44 gene predominantly distributed in coronary heart disease patients and cloned into pGL2-promoter and pGL2-control plasmids respectively containing luciferase gene, Hela cells and HepG2 cells were transfected. As a result, it was found that the expression level of recombinant plasmids inserted into the HVE44 allele in different cell lines was higher than that of the HVE36-inserted recombinant plasmids. These results suggest that the different alleles of the 3 ’hypervariable region of apolipoprotein B gene may play a role in regulating gene expression. HVE44 large allele may up-regulate intracellular gene expression, which may be dominated by large alleles such as HVE44 One of the causes of elevated apolipoprotein B in patients with atherosclerosis.
