目的:探讨红霉素对白介素(IL)-4诱导人支气管上皮细胞(HBE)表达IL-6及还原型谷胱甘肽(GSH)的影响。方法:培养HBE,取对数生长期细胞以每孔104个接种于96孔培养板中,分组为正常对照组(生理盐水20μL)、IL-4组(IL-410μL,生理盐水10μL)、红霉素低、高剂量组(IL-410μL,红霉素5、50μg.mL-110μL),加入相应药物,每组8个复孔,孵育24、48h后检测各组细胞存活情况及细胞中IL-6、GSH水平。结果:与正常对照组比较,IL-4组细胞孵育24、48h后活力明显降低(P<0.01),细胞中IL-6、GSH水平明显升高(P<0.05或P<0.01);与IL-4组比较,红霉素低、高剂量组细胞孵育48h后活力明显升高(P<0.05或P<0.01),细胞中IL-6、GSH水平明显降低(P<0.05或P<0.01),但孵育24h几乎无明显变化。结论:红霉素可抑制IL-4诱导HBE炎症介质IL-6的释放,下调GSH的合成。
Objective: To investigate the effect of erythromycin on the expression of IL-6 and glutathione (GSH) in human bronchial epithelial cells (HBE) induced by interleukin (IL) -4. Methods: The HBE was cultured and the cells in logarithmic growth phase were inoculated into 96-well plates at a rate of 104 per well. The cells were divided into normal control group (20μL normal saline group), IL-4 group (IL-410μL, normal saline 10μL) (IL-410μL, Erythromycin 5 and 50μg.mL-110μL) were added into the low-dose and high-dose groups, and the corresponding drugs were added into each group. Eight wells of each group were incubated for 24 and 48 hours. -6, GSH level. Results: Compared with the normal control group, the activity of IL-4 decreased significantly (P <0.01) and the levels of IL-6 and GSH in IL-4 group increased significantly (P <0.05 or P <0.01) (P <0.05 or P <0.01). The levels of IL-6 and GSH in the cells were significantly decreased (P <0.05 or P <0.01) , But almost no significant change after 24h incubation. Conclusion: Erythromycin can inhibit the release of IL-6 induced by IL-4 and down-regulate the synthesis of GSH.
