目的:利用糖尿病大鼠动物模型,通过观察大鼠视网膜中碱性成纤维细胞生长因子(basic fibroblast growth fac-tor,bFGF)蛋白表达、局部组织形态差异、晶状体等情况,探讨菩人丹在早期糖尿病视网膜病变(diabetic retinopathy,DR)治疗中的作用。方法:采用链脲佐菌素(strep tozocin,STZ)注入大鼠腹腔制造糖尿病大鼠动物模型。将实验大鼠随机分为正常组、模型组和治疗组,在5个月时处死动物取其视网膜,观察bFGF蛋白的表达情况、视网膜组织形态的改变情况、晶状体混浊程度以及血糖的变化情况,探讨菩人丹对糖尿病大鼠视网膜中bFGF蛋白表达的影响,以及视网膜组织形态学的改变情况。结果:在上述4个观察指标中,正常组大鼠均为阴性改变,模型组呈强阳性改变,治疗组大鼠呈弱阳性改变,差异有统计学意义(P<0.01)。结论:中药菩人丹在治疗早期糖尿病大鼠视网膜病变中可以有效抑制bFGF的表达,改善糖尿病大鼠视网膜血管的瘀血情况,轻度降低血糖,从而起到治疗DR的作用。
OBJECTIVE: To investigate the expression of basic fibroblast growth factor (bFGF) in retina of rats and the difference of local tissue morphology and lens in the animal model of diabetic rats. Role of diabetic retinopathy in the treatment of diabetic retinopathy. Methods: Animal model of diabetic rats was induced by intraperitoneal injection of streptozotocin (STZ). The rats were randomly divided into normal group, model group and treatment group. At 5 months, the animals were sacrificed and their retina was taken. The expression of bFGF protein, changes of retinal morphology, lens opacity and blood glucose were observed. To investigate the effect of Bodan Dan on bFGF protein expression in the retina of diabetic rats and the change of retinal histomorphology. Results: In the above four observation indexes, the normal rats were all negatively changed, the model group showed a strong positive change, the treatment group rats were weakly positive changes, the difference was statistically significant (P <0.01). Conclusion: Bodhidionan treatment can effectively inhibit the expression of bFGF, improve the blood stasis of retinal blood vessels in diabetic rats and slightly lower blood glucose in the treatment of diabetic retinopathy in early diabetic rats, and thus play a role in the treatment of DR.
