建立了高效液相色谱法同时测定盐酸达卡他韦及其5个光学异构体。采用Chiralpak IC色谱柱,以甲基叔丁醚(含0.1%二乙胺)∶甲醇(88∶12)为流动相,检测波长305 nm。盐酸达卡他韦及其5个光学异构体均分离完全,在0.1~4?g/ml范围内线性关系良好。方法回收率>99.0%,RSD<1.5%。
A high performance liquid chromatography (HPLC) was established for simultaneous determination of dakatavir hydrochloride and its five optical isomers. Using Chiralpak IC column with methyl tert-butyl ether (containing 0.1% diethylamine): methanol (88:12) as the mobile phase, detection wavelength of 305 nm. Dakatamivir hydrochloride and its five optical isomers were completely separated, with good linearity in the range of 0.1-4 μg / ml. Method recovery> 99.0%, RSD <1.5%.
