目的:了解哈尔滨市手足口病病原分布情况及基因特征,为哈尔滨市手足口病的预防控制工作提供参考。方法:收集哈尔滨市哨点医院的手足口病例样本,采用RT-PCR或real-time PCR方法进行病原核酸检测,病毒核酸阳性标本进行RD细胞培养分离病毒,将分离到的EV71采用RT-PCR的方法进行VP1区全基因扩增并测序,应用DNA Man和DNA Star软件序列整理分析。结果:2008年156份临床标本中EV71 59份,CA16 4份;2009年625份临床标本中EV71146份,CA16 82份,未分型别肠道病毒4份;2010年725份临床标本中EV71 202份,CA16 13份,未分型别肠道病毒36份。结论:通过基因比对2008年-2010年哈尔滨市EV71病毒感染手足口流行型别是EV71 C4亚型,未发现变异。
Objective: To understand the distribution and gene characteristics of HFMD in Harbin and to provide reference for the prevention and control of HFMD in Harbin. Methods: Samples of hand, foot and mouth were collected from sentinel hospital in Harbin. The pathogenic nucleic acid was detected by RT-PCR or real-time PCR. The virus nucleic acid positive samples were isolated and cultured in RD cells. The isolated EV71 was detected by RT-PCR Methods The full-length VP1 gene was amplified and sequenced. DNA Man and DNA Star software were used for sequence analysis. Results: In 2008, there were 59 EV71 and CA16 in 156 clinical samples. Among the 625 clinical samples in 2009, 7,116 were EV71, 82 were CA16, and 4 were not classified enterovirus. In 2010, 771 EV71 202 13 parts of CA16 and 36 parts of undifferentiated enterovirus. Conclusion: No genetic variation was found in EV71 C4 subtype of EV71 virus infection in hand, foot and mouth in Harbin from 2008 to 2010.
