目的探讨基质金属蛋白酶(MMP)-1、10、12、14及其组织抑制物——基质金属蛋白酶组织抑制物(TIMP)-3与女性压力性尿失禁(SUI)的关系。方法 2009年1月至2012年4月南京军区福州总医院收治并行手术治疗的64例SUI患者为观察组,分为未绝经组32例,绝经组32例,选取同期20例非压力性尿失禁患者做为对照组。采用逆转录聚合酶链反应(RT-PCR)检测尿道旁组织中MMP-1、10、12、14和TIMP-3的表达。结果观察组MMP-1和MMP-14 mR NA表达较对照组升高,差异有统计学意义(P<0.05);TIMP-3 mR NA表达较对照组明显降低,差异有统计学意义(P<0.01)。绝经组和未绝经组MMP-1、MMP-14和TIMP-3 mR NA表达差异无统计学意义(P>0.05)。3组MMP-10和MMP-12 mR NA表达差异无统计学意义(P>0.05)。结论增强的MMP-1和MMP-14的表达使盆底组织胶原纤维的降解增加;TIMP-3表达减弱,对胶原纤维降解的抑制作用减弱,可能参与压力性尿失禁的发生发展;但其表达与患者是否绝经无明显相关性。
Objective To investigate the relationship between matrix metalloproteinase (MMP) -1, 10, 12, 14 and its tissue inhibitor of matrix metalloproteinase tissue inhibitor (TIMP) -3 and female stress urinary incontinence (SUI). Methods From January 2009 to April 2012, 64 SUI patients who underwent concurrent surgery in Fuzhou General Hospital of Nanjing Military Region were enrolled in this study. There were 32 cases in the non-menopausal group and 32 cases in the menopause group. Twenty non-stress urinary incontinence Patients as a control group. Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of MMP-1, 10, 12, 14 and TIMP-3 in the peritumoral tissues. Results The expression of MMP-1 and MMP-14 mRNA in observation group was higher than that in control group (P <0.05), and the expression of TIMP-3 mR NA was significantly lower than that in control group (P < 0.01). The expression of MMP-1, MMP-14 and TIMP-3 mRNA in menopausal and non-menopausal groups had no significant difference (P> 0.05). There was no significant difference in the expression of MMP-10 and MMP-12 mR NA between the three groups (P> 0.05). Conclusions The enhanced expression of MMP-1 and MMP-14 may lead to the degradation of collagen fibers in pelvic floor. The expression of TIMP-3 is weakened and the inhibition of collagen fibers degradation is weakened, which may be involved in the development of stress urinary incontinence. However, No significant correlation with the patient’s menopause.
