目的探讨幼年肥胖鼠左室结构及功能变化时,心钠素(ANP)、内皮素(ET)在心肌组织中的表达变化以及血管紧张素Ⅱ受体拮抗剂洛沙坦的保护效应。方法复制实验性肥胖动物模型,将30只雄性大鼠随机分为对照组和高脂组,高脂组又分为用药组和非用药组,检测幼年肥胖鼠的左室质量(LVM)和左室质量指数(LVMI),应用放射免疫法测定肥胖幼鼠心肌组织中ANP、ET含量变化。结果①高脂非用药组与对照组相比,心肌组织中ANP、ET水平显著升高,差异有统计学意义(P<0.05),LVM、LVMI明显升高,差异有统计学意义(P<0.05);②经洛沙坦干预后,高脂用药组与高脂非用药组相比,心肌组织中ANP、ET表达水平明显下降,差异有统计学意义(P<0.05),LVM及LVMI明显降低,差异有统计学意义(P<0.05);③相关性分析表明,心肌组织中ANP、ET水平与LVM、LVMI呈正相关关系。结论心肌组织中ANP、ET含量变化及其相互关系在心肌肥厚的形成机制中可能起着重要的作用;小剂量洛沙坦能抑制幼年肥胖鼠心肌组织中ANP、ET的表达水平,从而逆转左室重构。
Objective To investigate the changes of left ventricular structure and function in juvenile obese rats, the changes of the expression of atrial natriuretic peptide (ANP) and endothelin (ET) and the protective effect of losartan, an angiotensin Ⅱ receptor antagonist. Methods Experimental animal models of obesity were reproduced. Thirty male rats were randomly divided into control group and hyperlipidemic group. The hyperlipidemic rats were divided into two groups: drug-treated group and non-drug-treated group. Left ventricular mass (LVM) The contents of ANP and ET in obese young rat myocardium were measured by radioimmunoassay. Results ① Compared with the control group, the levels of ANP and ET in the high-fat non-drug group were significantly increased (P <0.05), LVM and LVMI were significantly increased, the difference was statistically significant (P < 0.05). ② After intervention with losartan, the levels of ANP and ET in myocardial tissue in high-fat group were significantly lower than those in high-fat group (P <0.05), LVM and LVMI were significantly higher (P <0.05). (3) The correlation analysis showed that the levels of ANP and ET in myocardial tissue were positively correlated with LVM and LVMI. Conclusion The changes of myocardial ANP and ET levels and their correlation may play an important role in the mechanism of cardiac hypertrophy. Low-dose Losartan can inhibit the expression of ANP and ET in myocardial tissue of juvenile obese rats and reverse the left Room reconstruction.
