目的分离并鉴定我国首例输入性D9基因型麻疹病毒。方法使用Vero/SLAM细胞(非洲绿猴肾细胞/淋巴信号激活因子转染的非洲绿猴肾细胞)分离病毒,使用逆转录-聚合酶链反应扩增出编码核蛋白羧基末端450个核苷酸片段,通过对扩增产物进行核苷酸序列测定和分析,构建基因亲缘性关系树,进行遗传距离分析。结果四川省D9基因型麻疹病毒分离株MVi/Sichuan.CHN/07.09/1和世界卫生组织D9基因型代表株Victoria.AUS(维多利亚?澳大利亚)/12.99在基因亲缘性关系树上同属一个分支,核苷酸同源性为96.9%;和2008年流行于泰国和荷兰的D9基因型麻疹野病毒的核苷酸和氨基酸同源性分别为99.8%~100%和99.3%~100%;和中国大陆目前所使用的麻疹疫苗株沪191相比对,其核苷酸和氨基酸同源性分别为92.3%和90.7%;和中国目前流行的麻疹病毒绝对优势本土基因型H1基因型代表株相比对,其核苷酸和氨基酸同源性分别为90.8%和92.1%。结论输入中国四川省的麻疹病毒株为D9基因型。
Objective To isolate and identify the first imported D9 genotype measles virus in China. The nucleotide and amino acid homology of wild type D9 genotype wild virus in Thailand and the Netherlands in 2008 were 99.8% -100% and 99.3% -100%, respectively; The presently used measles vaccine strain Shanghai 191 compared to its nucleotide and amino acid homologies were 92.3% and 90.7%; and China’s current epidemic of measles virus absolute advantage native genotypes H1 genotype representative strains compared to The nucleotide and amino acid homologies were 90.8% and 92.1% respectively. Conclusion The measles virus strain imported into Sichuan Province of China was D9 genotype.
