目的:探讨7-二氟甲基金雀异黄素对PC12细胞氧化应激损伤的保护作用。方法:采用过氧化氢(H2O2)诱导PC12细胞氧化应激损伤的模型,MTT法检测细胞增殖活性,碘化丙啶(PI)染色流式细胞术(FCM)检测细胞凋亡,多功能生化分析仪测定培养液上清中的乳酸脱氢酶(LDH)活性。结果:7-二氟甲基金雀异黄素能有效拮抗40.0μmol/L的H2O2孵育24h对PC12细胞增殖活性的抑制作用和对PC12细胞凋亡的诱导作用,并能减少40.0μmol/L的H2O2诱导的PC12细胞的LDH释放。结论:7-二氟甲基金雀异黄素对过氧化氢诱导PC12细胞的氧化应激损伤具有保护作用。
Objective: To investigate the protective effect of 7-difluoromethyl genistein on oxidative stress injury in PC12 cells. Methods: H2O2 was used to induce oxidative stress injury in PC12 cells. Cell proliferation was detected by MTT assay. Cell apoptosis was detected by flow cytometry (FCM) with propidium iodide (PI) staining and multifunctional biochemical analysis Determination of lactate dehydrogenase (LDH) activity in the culture supernatant. Results: 7 - Difluoromethylgenin effectively antagonized the inhibitory effect of 40.0μmol / L H2O2 for 24h on the proliferation of PC12 cells and the induction of apoptosis in PC12 cells and decreased the induction of 40.0μmol / L of H2O2 LDH release from PC12 cells. CONCLUSION: 7-Difluoromethylgenisin has a protective effect on oxidative stress injury induced by hydrogen peroxide in PC12 cells.
