本文用微核试验的方法检测了新疆产莫合烟(一种组制烟草)对小鼠胚胎细胞的遗传毒作用,并与新疆卷烟厂生产的天池牌香烟加以比较。给妊娠13─15d的瑞士小鼠腹腔注射烟雾凝聚物,共二次,间隔24h,第二次染毒后16h杀死动物,取胎鼠的肝脏及母鼠的骨髓,制片,计数多染红细胞(PCE)的微核率。结果表明莫合烟烟雾凝聚物在中等浓度(120mg/kg)时胎肝细胞的微核率(7.93±2.23)与溶剂对照(6.17±1.85)相比有显著性差异。骨髓细胞的微核率(8.31±2.21)与对照细(5.11±1.05)相比,亦有显著增高。在高浓度(240mg/kg)时,胎肝细胞的微核率上升至12.13±4.43.骨髓细胞为10.67±4.33,与溶剂对照相比有极显著差异(P<0.0005)。且均呈剂量效应关系。而天池香烟只在高浓度时胎肝细胞微核率(9.29±2.56)与骨髓细胞微核率(5.5±2.07)的增高才有显著意义。两种烟相比较,在高涨度即1.3支烟/kg时,莫合烟诱发的胎肝细胞微核率大于天池香烟(P<0.05)。以上结果说明莫合烟雾凝聚物对小鼠胚胎细胞有遗传毒性,其毒性在高浓度时大于天池香烟。
In this paper, we examined the genetic toxic effects of MOF (a kind of tobacco made in China) on mouse embryonic cells by means of micronucleus test and compared with Tianchi cigarettes produced by Xinjiang Tobacco Factory. Thirteen to fifteen days of gestation, Swiss mice were injected intraperitoneally with smoke condensate twice a day for 24 hours. Animals were sacrificed 16 h after the second exposure, and the liver and fetal bone marrow of fetuses were taken and counted. Micronucleus rate of erythrocytes (PCE). The results showed that the micronucleus rate (7.93 ± 2.23) and the solvent control (6.17 ± 1.85) of the MOF smog coagulant at the medium concentration (120 mg / kg) were significant difference. The micronucleus rate of bone marrow cells (8.31 ± 2.21) was also significantly higher than the control (5.11 ± 1.05). At high concentrations (240 mg / kg), the micronucleus rate of fetal liver cells increased to 12.13 ± 4.43. Bone marrow cells were 10.67 ± 4.33, which was significantly different from the solvent control (P <0.0005). And were dose-response relationship. However, Tianchi cigarettes had significant significance only at high concentration of fetal liver micronucleus rate (9.29 ± 2.56) and bone marrow micronucleus rate (5.5 ± 2.07). Compared with the two cigarettes, the rate of micronuclei of fetal liver cells induced by MOF was higher than that of Tianchi cigarettes (P <0.05) at the high degree of 1.3 cigarettes / kg. The above results show that Mohs smoke condensate has genotoxicity on mouse embryonic cells and its toxicity is higher than that of Tianchi cigarettes at high concentration.
