目的应用蛋白质组学技术筛选人胆汁中与胆管癌相关的蛋白质分子。方法收集2008年3月至2009年10月重庆医科大学附属第一医院肝胆外科经明确病理诊断的胆管癌住院患者的胆汁共15例,正常人胆汁5例。应用非离子型脱脂剂结合分子筛超滤方法制备纯化胆汁样本,通过高效液相色谱-质谱法(HPLC-CHIP-MS/MS)鉴定差异表达的蛋白质,并用Spectrum Mill software对检测结果进行分析。结果与正常人胆汁比较,在胆管癌患者胆汁中,检测到109个高可信蛋白,其中12个为新增蛋白质,其功能主要涉及癌细胞的发生、增殖、分化、转移等。结论高效液相色谱-质谱法鉴定的12个差异表达蛋白质可能成为研究胆管癌的生物学分子标志物。
Objective To screen proteoglycan associated with cholangiocarcinoma in human bile by proteomics. Methods A total of 15 patients with cholangiocarcinoma inpatients admitted to the First Affiliated Hospital of Chongqing Medical University from March 2008 to October 2009 were collected. The bile samples were collected from 5 patients with normal bile. Purified bile samples were prepared by non-ionic degreasing agent combined with molecular sieve ultrafiltration. The differentially expressed proteins were identified by HPLC-CHIP-MS / MS and analyzed by Spectrum Mill software. Results Compared with normal bile, 109 high-credible proteins were detected in bile of cholangiocarcinoma, of which 12 were newly added proteins. Their functions mainly involved in the occurrence, proliferation, differentiation and metastasis of cancer cells. Conclusion The 12 differentially expressed proteins identified by high performance liquid chromatography-mass spectrometry may become biological molecular markers for the study of cholangiocarcinoma.
