目的:探讨滋补脾阴方药(Zibu Piyin Recipe,ZBPYR)对大鼠脊髓损伤(spinal cord injury,SCI)的治疗效果以及对低氧诱导因子(hypoxia inducible factor,HIF)-1α的表达的影响。方法:将清洁级Wistar大鼠36只随机分为假手术组、阳性对照组(喜得镇组,0.6 mg.kg-1)、SCI组和ZBPYR低、中、高剂量组(分别为7,14,21 g.kg-1)。各组大鼠分别于伤后72 h处死,并用Western-blot和RT-PCR方法检测HIF-1α和HIF-1αmRNA的表达。结果:伤后72 h,HIF-1α和HIF-1αmRNA在SCI组的表达量显著高于假手术组、ZBPYR低、中、高剂量组和阳性对照组,差异具有统计学意义(P<0.05);HIF-1α和HIF-1αmRNA的表达在ZBPYR低、中、高剂量组间的差异具有统计学意义(P<0.05)。结论:ZBPYR可改善受损脊髓局部微环境的缺血缺氧程度来抑制SCI后继发性损伤的发生和发展,其作用效果具有浓度依赖性。
Objective: To investigate the therapeutic effect of Zibu Piyin Recipe (ZBPYR) on spinal cord injury (SCI) and the expression of hypoxia inducible factor (HIF) -1α in rats. Methods: Thirty-six Wistar rats were randomly divided into sham operation group, positive control group (0.6 mg.kg-1), SCI group and ZBPYR low, middle and high dose groups (7, 14, 21 g.kg-1). The rats in each group were killed at 72 h after injury, and the expressions of HIF-1α and HIF-1αmRNA were detected by Western-blot and RT-PCR. Results: The expression of HIF-1α and HIF-1αmRNA in SCI group was significantly higher than that in sham operation group, ZBPYR low, medium and high dose group and positive control group at 72 h after injury (P <0.05) ; The expression of HIF-1α and HIF-1αmRNA in ZBPYR low, medium and high dose groups had statistical significance (P <0.05). Conclusion: ZBPYR can improve the degree of hypoxia and hypoxia in the local microenvironment of injured spinal cord to inhibit the occurrence and development of secondary injury after SCI. The effect of ZBPYR in a concentration-dependent manner.
