目的:探求ALA口服联合小剂量HPD注射在鼠脑组织及脑肿瘤中的代谢,寻求静脉注射小剂量HPD联合口服ALA脑肿瘤光敏诊断及治疗的最佳的ALA口服剂量及时间。方法:鼠单纯静脉注射1.25mg/kg、口服不同剂量的ALA(20mg/kg、40mg/kg、60mg/kg),及二者联合,及在不同时间(用药前、用药后1小时、2小时、4小时、6小时、8小时、10小时、12小时、24小时、48小时),以激光诱导荧光OMA检测系统测定脑肿瘤及正常脑组织荧光,并与未用药健康鼠脑组织,及未用药脑肿瘤鼠脑组织作对比,并经数据处理,寻求脑肿瘤与正常脑组织荧光相对值相差最大的合适的ALA口服剂量及诊断时间。结果:单纯用1.25mg/kg的HPD在用药后8小时脑肿瘤组织与正常脑组织630nm除以610荧光峰相对值之比值最大,单纯用口服ALA20mg/kg在口服后8小时比值最大,单纯口服ALAALA40-60mg/kg,比值4-6小时比值最大。静脉注射1.25mg/kg的HPD合并口服ALA20mg/kg(简称M20组)在用药后8小时比值最大,合并口服ALA40-60mg/kg组(简称M40、M60组)比值4-6小时比值最大。结论:静脉注射HPD1.25mg/kg合并口服ALA20mg/kg(简称M20组),该小剂量使用光敏剂同样能达到光敏诊断及治疗目的。鼠在用药后8小时作光敏诊断及光动力学治疗最合适,而人在用药后24小时是最佳的光敏诊断及治疗的最佳时间。
OBJECTIVE: To explore the optimal ALA oral dose and time for ALA oral administration combined with low-dose HPD injection in brain tissue and brain tumor of mice and to seek intravenous low-dose HPD combined with oral ALA brain tumor for light-sensitive diagnosis and treatment. Methods: The rats were given intravenous injection of 1.25 mg / kg of ALA (20 mg / kg, 40 mg / kg, 60 mg / kg) at different doses and the combination of the two at different times (before treatment, 1 hour and 2 hours , 4 hours, 6 hours, 8 hours, 10 hours, 12 hours, 24 hours, 48 hours). The laser-induced fluorescence OMA detection system was used to measure the fluorescence of brain tumor and normal brain tissue. The brain tissues of mice with brain tumor were compared with each other. After data processing, the suitable ALA oral dose and diagnosis time with the largest difference between the relative fluorescence values of brain tumor and normal brain tissue were obtained. Results: The ratio of 630nm divided by 610 fluorescence peak was the highest in pure brain tissue of HPD with 1.25mg / kg HPD at 8h after treatment, and the highest ratio was obtained at 8h after oral administration of oral ALA alone. ALAALA 40-60mg / kg, the ratio of 4-6 hours the ratio of the largest. Intravenous 1.25mg / kg HPD combined with oral ALA20mg / kg (referred to as the M20 group) the ratio of 8 hours after treatment the largest, combined with oral ALA40-60mg / kg group (referred to as M40, M60 group) ratio 4-6 hours the ratio. Conclusion: Intravenous injection of HPD1.25mg / kg combined oral ALA20mg / kg (referred to as M20 group), the small dose of photosensitizer can also achieve the purpose of photosensitive diagnosis and treatment. Rats in the 8 hours after treatment for photodynamic diagnosis and photodynamic therapy is the most appropriate, and 24 hours after treatment is the best time for photosensitive diagnosis and treatment of the best.
