目的探讨胰岛素样生长因子Ⅱ(IGF-Ⅱ)在血管瘤增殖与退化中的作用。方法应用免疫组化方法检测52例血管瘤、27例血管畸形和5例正常皮肤标本中IGF-Ⅱ及细胞增殖核抗原Ki-67的表达情况。同时应用RT-PCR法检测9例血管瘤、5例血管畸形和5例正常皮肤标本中IGF-ⅡmRNA的表达情况。结果免疫组化结果显示血管瘤组的IGF-Ⅱ和Ki-67表达均高于血管畸形组及正常皮肤组(P<0.01)。血管瘤组中增生期亚组两标记指数均高于退化期亚组(P<0.01)。血管瘤组中IGF-Ⅱ与Ki- 67标记指数的相关系数为0.84,P<0.01。RT-PCR法检测到增殖期血管瘤中有明显IGF-ⅡmRNA的表达,而消退期血管瘤、血管畸形和正常皮肤中未见明显IGF-ⅡtuRNA的表达。结论IGF-Ⅱ与血管瘤内皮细胞的增殖相关。
Objective To investigate the role of insulin-like growth factor Ⅱ (IGF-Ⅱ) in the proliferation and degeneration of hemangiomas. Methods Immunohistochemistry was used to detect the expression of IGF-Ⅱ and Ki-67 in 52 hemangiomas, 27 vascular anomalies and 5 normal skin specimens. Meanwhile, the expression of IGF-ⅡmRNA in 9 hemangiomas, 5 vascular malformations and 5 normal skin specimens was detected by RT-PCR. Results The results of immunohistochemistry showed that the expression of IGF-Ⅱ and Ki-67 in hemangioma group were higher than that in vascular malformation group and normal skin group (P <0.01). Both markers in hyperplasia subgroups in hemangiomas were higher than those in degenerative subgroups (P <0.01). The correlation coefficient of IGF-Ⅱ and Ki- 67 labeling index in hemangioma group was 0.84, P <0.01. The expression of IGF-ⅡmRNA in proliferating hemangiomas was detected by RT-PCR, while the expression of IGF-ⅡtuRNA in retching hemangiomas, vascular malformations and normal skin was not observed. Conclusion IGF-Ⅱ is related to the proliferation of hemangiomas endothelial cells.
