目的探讨盐酸地尔硫卓对癫痫幼鼠海马中多药耐药(MDR)基因表达的影响。方法将75只出生7 d Wistar大鼠随机分为海人酸(KA)1 mg/kg组(A组,39只)和对照组(B组,36只)。将造模成功的A组31只大鼠随机分为A1(10只)、A2(11只)和A3组(10只),B组随机均分为B1、B2和B3组。其中,A2、B2组予拉莫三嗪治疗,A3、B3组予拉莫三嗪及盐酸地尔硫卓联合治疗。用药6周后,将各组大鼠断头取海马,用RT-PCR法测定mdr1a和mdr1b mRNA表达。结果与B1组相比,A1、A2组大鼠海马中mdr1a和mdr1b mRNA表达明显升高(P<0.01),而A3组mdr1a和mdr1b mRNA表达低于A1、A2组(P<0.05);B3组mdr1a和mdr1b mRNA表达与B1、B2组无统计学差异(P>0.05)。结论盐酸地尔硫卓可以抑制癫痫幼鼠海马中MDR基因表达的增加。
Objective To investigate the effect of diltiazem hydrochloride on the multidrug resistance (MDR) gene expression in the hippocampus of epileptic young rats. Methods Seventy-five Wistar rats were randomly divided into 3 groups: KA 1 mg / kg (group A, n = 39) and control group (group B, n = 36). A total of 31 rats in group A were randomly divided into A1 (n = 10), A2 (n = 11) and A3 (n = 10). Group B was randomly divided into B1, B2 and B3 groups. Among them, A2, B2 group lamotrigine treatment, A3, B3 group lamotrigine hydrochloride and diltiazem hydrochloride combination therapy. After 6 weeks of treatment, the rats in each group were decapitated and the expression of mdr1a and mdr1b mRNA was determined by RT-PCR. Results Compared with B1 group, the expression of mdr1a and mdr1b mRNA in the hippocampus of rats in A1 and A2 groups was significantly increased (P <0.01), while the mRNA expression of mdr1a and mdr1b in A3 group was lower than that in A1 and A2 groups (P <0.05) The mRNA expression of mdr1a and mdr1b in group B1 and B2 had no statistical difference (P> 0.05). Conclusion Diltiazem hydrochloride can inhibit the increase of MDR gene expression in the hippocampus of epileptic young rats.
