目的观察不同发育阶段家兔直肠内胰岛淀粉样多肽(islet amyloid polypeptide,IAPP)、P物质(substance P,SP)的表达及分布特点。方法收集生后5、15、25、35、60 d及90 d家兔各10只。取直肠组织入Bouin液固定,梯度酒精脱水,常规石蜡包埋,制成连续切片。用免疫组织化学SABC法和图像分析法对IAPP、SP免疫反应阳性细胞的定位和表达进行观察。结果生后5 d家兔直肠内可见少量散在分布IAPP阳性细胞,多位于上皮;35 d IAPP阳性细胞的数量呈明显上升趋势,至60 d达到最多(P<0.05),90 d时减少(P<0.05),主要分布于固有层。图像分析测得IAPP阳性细胞的平均灰度值随家兔的生长发育逐渐降低,60 d达最低(P<0.05)。SP阳性细胞于生后15 d始出现,35 d后细胞数量明显增多(P<0.05),60 d达最高(P<0.05)。其平均灰度值也于35 d开始下降(P<0.05),于60 d达最低水平(P<0.05)。结论 IAPP、SP在直肠的表达随家兔的生长发育而逐渐变化,推测两者参与调节生后发育期间家兔直肠消化功能的成熟过程。
Objective To observe the expression and distribution of islet amyloid polypeptide (IAPP) and substance P (SP) in rabbits at different developmental stages. Methods Totally 10 rabbits were collected at 5, 15, 25, 35, 60 d and 90 d after birth. Take rectal tissue into the Bouin solution, gradient alcohol dehydration, paraffin embedded, made into serial sections. The localization and expression of IAPP and SP immunoreactive cells were observed by immunohistochemical SABC method and image analysis. Results A small amount of scattered IAPP positive cells were observed in the rectum of rabbits 5 days after birth. The numbers of IAPP positive cells in the rectum increased significantly on the 35th day, reaching the peak at 60 days (P <0.05) and decreasing at 90 days (P <0.05), mainly distributed in the lamina propria. The average gray value of IAPP positive cells decreased gradually with the growth and development of rabbits and reached the lowest level at 60 d (P <0.05). The number of SP positive cells appeared at 15 days after birth, and the number of cells increased significantly after 35 days (P <0.05), reaching the highest on the 60th day (P <0.05). The mean gray value also decreased at 35 days (P <0.05) and reached the lowest level at 60 days (P <0.05). Conclusion The expressions of IAPP and SP in the rectum gradually change with the growth and development of rabbits. It is speculated that both of them are involved in the regulation of the rectal digestive function during the development of the rabbit.
