目的:定量检测细胞周期素E(Cyclin E)在喉鳞状细胞癌及颈部淋巴组织中的表达,探讨其在喉癌转移中的可能作用。方法:采用激光共聚焦显微镜、多参数流式细胞技术对手术切除的21例喉鳞状细胞癌(7例伴颈淋巴结转移,14例不伴颈淋巴结转移)组织标本中Cyclin E的表达含量进行检测分析。结果:Cyclin E在喉鳞状细胞癌、阳性淋巴结、阴性淋巴结中均有不同程度的表达,喉鳞状细胞癌伴颈淋巴结转移平均荧光强度为78.12±6.54,略高于不伴颈淋巴结转移者(74.93±8.82)(P>0.05);阳性淋巴结平均荧光强度为93.65±7.48,明显高于阴性淋巴结(30.65±7.48)(P<0.01),且高于喉鳞状细胞癌伴颈淋巴结转移和不伴颈淋巴结转移者(P<0.05)。结论:Cyclin E的异常表达是喉癌发生、发展过程中一个重要的分子事件;Cyclin E的检测可望成为临床评估喉癌淋巴结转移的指标之一。
Objective: To quantitatively detect the expression of Cyclin E in laryngeal squamous cell carcinoma and cervical lymphoid tissue, and to explore its possible role in the metastasis of laryngeal carcinoma. Methods: The expression of Cyclin E in 21 specimens of laryngeal squamous cell carcinoma (7 cases with cervical lymph node metastasis and 14 cases without cervical lymph node metastasis) was examined by laser confocal microscopy and multi-parameter flow cytometry Test analysis. Results: The expression of Cyclin E in laryngeal squamous cell carcinoma, positive lymph nodes and negative lymph nodes were all different. The average fluorescence intensity of laryngeal squamous cell carcinoma with cervical lymph node metastasis was 78.12 ± 6.54, which was slightly higher than those without cervical lymph node metastasis (74.93 ± 8.82) (P> 0.05). The average fluorescence intensity of positive lymph nodes was 93.65 ± 7.48, which was significantly higher than that of negative lymph nodes (30.65 ± 7.48) (P <0.01), which was higher than that of laryngeal squamous cell carcinoma with cervical lymph node metastasis Not accompanied by cervical lymph node metastasis (P <0.05). Conclusion: The abnormal expression of Cyclin E is an important molecular event in the development and progression of laryngeal carcinoma. The detection of Cyclin E is expected to become one of the clinical indexes for lymph node metastasis of laryngeal cancer.
