目的:观察天麻素对离体大鼠心肌缺血/再灌注(I/R)损伤的影响。方法:应用Langendorff离体心脏灌注系统建立心肌缺血/再灌注损伤模型。40只SD大鼠随机分为正常对照组、缺血/再灌注组、天麻素0.1、0.025mmol·L-1组。除正常对照组外,各组分别平衡灌注、全心停灌和再灌注各30min,观察天麻素对左室发展压(LVDP)、冠状动脉流量(CF)、心肌组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量以及超微结构的影响。结果:天麻素明显抑制I/R引起的LVDP和CF的下降,显著改善缺血/再灌注后的心功能,且有明显的量效关系。提高心肌SOD活力和降低MDA含量。显著减轻I/R引起的心肌超微结构的损伤性改变。结论:天麻素对I/R引起的心肌损伤有显著的拮抗作用,其机制可能与其增强清除自由基能力,减轻生物膜脂质过氧化反应,维持心肌细胞膜和线粒体的正常功能,改善能量代谢有关。
Objective: To observe the effects of gastrodin on myocardial ischemia/reperfusion (I/R) injury in isolated rat. METHODS: A rat model of myocardial ischemia/reperfusion injury was established using a Langendorff heart perfusion system. Forty SD rats were randomly divided into normal control group, ischemia/reperfusion group, and gastrodin 0.1, 0.025 mmol·L-1 group. In addition to the normal control group, the groups were equilibrated with perfusion, cardiotonic arrest, and reperfusion for 30 min each, and the effects of gastrodin on left ventricular development pressure (LVDP), coronary artery flow (CF), and myocardial tissue superoxide dismutase (SOD) were observed. ) Viability and malondialdehyde (MDA) content and ultrastructure effects. RESULTS: Gastrodin significantly inhibited the decrease of LVDP and CF induced by I/R, significantly improved cardiac function after ischemia/reperfusion, and showed a significant dose-effect relationship. Improve myocardial SOD activity and reduce MDA content. Significantly reduce I/R-induced changes in myocardial ultrastructural damage. Conclusion: Gastrodin has a significant antagonistic effect on I/R-induced myocardial injury. Its mechanism may be related to its ability to enhance free radical scavenging, reduce lipid peroxidation of biofilm, maintain normal function of myocardial cell membrane and mitochondria, and improve energy metabolism. .
