目的通过临床对甲状腺癌患者采用逆转录聚合酶链反应(RT-PCR)检测方法对患者的外周血中促甲状腺素受体mRNA(TSHR-mRNA)和甲状腺球蛋白mRNA(Tg-mRNA)进行检测,分析其在甲状腺癌中的表达,并研究其检测结果对甲状腺癌的诊断意义。方法研究对象均选自哈尔滨医科大学第二医院甲状腺外科住院患者,共170例,分正常对照组(30例);甲状腺良性疾病组(40例);甲状腺癌组(100例)。对三组患者分别采用RT-PCR检测方法,比较三组患者的Tg-mRNA、TSHR-mRNA的水平以及临床表达,分析TSHR-mRNA和Tg-mRNA在甲状腺癌中的诊断意义。结果甲状腺癌组患者的TSHR-mRNA以及Tg-mRNA阳性率明显高于正常对照组和甲状腺良性疾病组,差异有统计学意义(P<0.05)。正常甲状腺疾病患者的TSHR-mRNA以及Tg-mRNA水平变化与正常对照组差异无备统计学意义(P>0.05)。结论促TSHR-mRNA以及Tg-mRNA能够准确的反映甲状腺患者的病情。
Objective To detect thyrotropin receptor mRNA (TSHR-mRNA) and thyroglobulin mRNA (Tg-mRNA) in peripheral blood of patients with thyroid cancer by reverse transcription polymerase chain reaction (RT-PCR) , Analyze its expression in thyroid cancer, and study the diagnostic value of its test results for thyroid cancer. Methods The subjects were all from 170 patients with thyroid surgery in the Second Hospital of Harbin Medical University. They were divided into normal control group (30 cases), thyroid benign disease group (40 cases) and thyroid cancer group (100 cases). The diagnostic value of TSHR-mRNA and Tg-mRNA in thyroid cancer was analyzed by RT-PCR, and the levels of Tg-mRNA and TSHR-mRNA were compared among the three groups. Results The positive rates of TSHR-mRNA and Tg-mRNA in thyroid cancer patients were significantly higher than those in normal control group and benign thyroid disease group (P <0.05). TSHR-mRNA and Tg-mRNA levels in patients with normal thyroid disease were not significantly different from those in the control group (P> 0.05). Conclusions Promoting TSHR-mRNA and Tg-mRNA can accurately reflect the condition of thyroid patients.
