组胺释放因子(HRF)是一组细胞因子,由单核细胞等多种细胞在抗原或丝裂原刺激下产生。HRF 可导致嗜硷细胞脱颗粒并释放组胺。巳经证明HRF 与支气管的超反应性密切相关,而这种气道的超反应性正是哮喘的特征。因此,作者设计本实验以弄清呼吸道病毒能否在体外诱导人单核细胞产生HRF。实验方法:1.取健康、非过敏体质的成人外周血,经ficoll-hypague 沉淀技术获得单核细胞,其中含有1%~3%的嗜硷细胞,并作细胞计数,将5×10~6/ml 细胞分别加入预先制备的含有流感病毒A 和含有呼吸道合胞病毒的悬液以及对照介质中,置37℃下孵化2小时,然后将细胞洗涤3次,洗涤后的细胞重悬于增添10%供者自身血浆的培养
