目的通过体外原代培养人大网膜前脂肪细胞,并诱导其分化至成熟,研究雄激素对脂肪细胞分泌瘦素的影响。方法选取择期开腹手术患者的新鲜脂肪组织,采用胶原酶消化的方法分离培养人大网膜前脂肪细胞,诱导分化至成熟后分别用1、10、100nmol/L的睾酮干预24h后收集上清液。用酶联免疫吸附法同批检测所收集细胞上清液中瘦素蛋白含量。结果100nmol/L的睾酮对已分化前脂肪细胞分泌的瘦素有抑制作用。结论成功建立了原代人网膜前脂肪细胞体外培养模型,通过观察雄激素对脂肪细胞分泌瘦素的影响,为研究肥胖及相关代谢异常疾病的性别差异提供一些实验室证据。
Objective To study the effect of androgens on the secretion of leptin in adipocytes by primary culture of human omental preadipocytes in vitro and to induce their differentiation to maturation. Methods Fresh adipose tissue from patients undergoing elective laparotomy was isolated and cultured in human omental preadipocytes by collagenase digestion. After differentiation and maturation, the supernatants were harvested with 1, 10 and 100 nmol / L of testosterone for 24 h, respectively. . Enzyme-linked immunosorbent assay with the same batch of leptin protein content in the supernatant collected cells. Results 100nmol / L of testosterone can inhibit the secretion of leptin from preadipocytes. Conclusion The primary culture model of prolactin preadipocytes was established successfully. By observing the effect of androgen on the secretion of leptin by adipocytes, we provided some laboratory evidence for studying the gender differences of obesity and related metabolic disorders.
