目的:探讨胰岛素样生长因子Ⅰ受体α链靶向阻断对人肺腺癌细胞生长的影响。方法:应用杂交瘤技术制备IGFⅠ-αR单克隆抗体,经ProteinG柱纯化分离单克隆抗体;MTT法检测细胞的增殖情况。结果:IGFⅠ-αR单克隆抗体明显抑制IGFⅠ与细胞膜表面IGFⅠ-αR结合能力,抗体浓度在150ng/ml时抑制率达到90%;随着抗体浓度的降低,其抑制率相应下降,存在明显的剂量-效应关系。IGFⅠ-αR单克隆抗体干预后,SPC-A-1、A549两组细胞均增长缓慢,分别与相应空白对照组比较,具有显著性差异(P<0.05〉;但两组间差异不显著。结论:本研究提取的IGFⅠ-αR单克隆抗体对IGFⅠ-αR具有较好亲和力,能明显抑制SPC-A-1、A549肺腺癌细胞株增殖。
OBJECTIVE: To investigate the effect of targeted blockade of insulin-like growth factor I receptor alpha on the growth of human lung adenocarcinoma cells. Methods: Monoclonal antibody of IGF Ⅰ-αR was prepared by hybridoma technique. Monoclonal antibody was purified by ProteinG column. The proliferation of cells was detected by MTT assay. RESULTS: Monoclonal antibodies against IGFⅠ-αR significantly inhibited the binding of IGFⅠ to IGFⅠ-αR on the cell membrane surface. The inhibitory rate reached 90% when the antibody concentration was 150ng / ml. With the decrease of antibody concentration, the inhibitory rate decreased obviously. - the relationship between effects. After the intervention of IGFⅠ-αR monoclonal antibody, the cells in both SPC-A-1 and A549 groups grew slowly, which were significantly different from the corresponding blank control group (P <0.05), but there was no significant difference between the two groups. The IGFⅠ-αR monoclonal antibody extracted in this study has good affinity to IGFⅠ-αR, and can significantly inhibit the proliferation of SPC-A-1 and A549 lung adenocarcinoma cell lines.
