目的 探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)基因对体内转染皮瓣成活的影响。方法 将30只SD大鼠随机均分为pcDNA_3-VEGF_(165)组、pcDNA_3组(两组均为术前48h、术中两次转染)和生理盐水(对照)组,术后7d计算各组大鼠皮瓣的成活面积。对皮肤标本行病理观察,免疫组化染色检测VEGF表达情况。结果 三组大鼠术后皮瓣平均成活率分别为48.46+3.35%、30.20±2.16%、31.35±1.99%,pcDNA_3-VEGF_(165)组显著高于其余两组(P<0.05);免疫组化染色示pcDNA_3-VEGF_(165)组染色深度亦显著深于其余两组(P<0.05)。结论 转染pcDNA_3-VEGF_(165)能够改善任意皮瓣的成活率,并显著表达VEGF_(165)。
Objective To investigate the effect of vascular endothelial growth factor (VEGF) gene on the survival of transfected flaps in vivo. Methods Thirty Sprague-Dawley rats were randomly divided into two groups: pcDNA3-VEGF16 group and pcDNA3 group (48h, twice transfection during operation) and saline group (control group) Group rat flap survival area. Pathological examination of skin specimens, immunohistochemical staining VEGF expression. Results The average survival rate of the flaps in the three groups were 48.46 ± 3.35%, 30.20 ± 2.16% and 31.35 ± 1.99%, respectively. The pcDNA3-VEGF165 group was significantly higher than the other two groups (P <0.05) The staining depth of pcDNA3-VEGF165 group was also significantly deeper than the other two groups (P <0.05). Conclusion Transfection of pcDNA_3-VEGF_ (165) can improve the survival rate of any skin flap and express VEGF_ (165).
