研究了合成酸性磷酸酶(ACP)检测底物2,6-二氯-4-乙酰基苯基磷酸(DCAP-P)的新方法。以2,6-二氯苯酚为原料,经酰化反应、AlCl3催化F-C重排先制得2,6-二氯-4-乙酰基苯酚(DCAP),然后在低温下向含有DCAP和三氯氧磷的甲苯溶液中滴加吡啶将其磷脂化,再经水解就得到了目标产物DCAP-P,其结构经元素分析、1HNMR和IR得到确证。研究表明该方法具有步骤较少、操作简便、产物易分离纯化和收率良好等优点。
A new method of synthesizing acid phosphatase (ACP) for detecting substrate 2,6-dichloro-4-acetylphenyl phosphate (DCAP-P) was studied. Using 2,6-dichlorophenol as raw material, 2,6-dichloro-4-acetylphenol (DCAP) was prepared by acylation reaction and AlCl3-catalyzed FC rearrangement. Then, DCAP was treated with DCAP and trichlorooxy Phosphorus toluene solution was added dropwise to the phospholipid of pyridine, and then hydrolyzed to give the target product DCAP-P, its structure was confirmed by elemental analysis, 1HNMR and IR. The research shows that the method has the advantages of fewer steps, simple and convenient operation, easy separation and purification, good yield and so on.
