目的:研究周围神经65KD蛋白在损伤性大鼠坐骨神经中的分布与表达。方法:取损伤15d后的大鼠坐骨神经近侧端及远侧端,冰冻切片,与抗65KD蛋白单克隆抗体与切片孵育后,加羊抗小鼠IgGHRP,DAB反应成色。结果:大鼠坐骨神经远侧端及近侧端中均有65KD蛋白阳性免疫反应产物,阳性免疫反应产物位于雪旺氏细胞。阳性免疫反应产物的密度及强度在神经的远侧端大于近侧端。结论:神经诱向因子65KD蛋白由雪旺氏细胞产生。在损伤后神经近、远侧端都存在65KD蛋白,远侧端的强度大于近侧端,这一浓度梯度可能是诱导再生的神经纤维定向生长过程中的一个重要因素。
Objective: To study the distribution and expression of 65KD protein in the sciatic nerve of injured rats. Methods: The sciatic nerve of proximal and distal sciatic nerve of rats after 15 days of injury were frozen and sectioned. After incubated with anti-65KD monoclonal antibody and sections, goat anti-mouse IgG-HRP and DAB reaction were performed. Results: The rat sciatic nerve distal and proximal ends of both 65KD protein-positive immune reaction products, the positive immune reaction products located in Schwann cells. The density and intensity of positive immune reaction products are greater at the distal end of the nerve than at the proximal end. Conclusion: The 65 kD neurotransmitter protein is produced by Schwann cells. After injury, there are 65KD proteins in the proximal and distal ends of the nerve, and the strength of the distal end is greater than that of the proximal end. This concentration gradient may be an important factor in the directional growth of nerve fibers that induce regeneration.
