目的:分析IL4对亚急性重型肝炎(亚重肝)患者外周血单个核细胞(PBMCs)TNFα和IL1的调控作用,评价IL4在亚重肝治疗中的潜在价值。方法:应用细胞生物学、免疫组化和RTPCR等方法测定PBMCsTNFα、IL1的表达。结果:亚重肝患者PBMCsTNFα、IL1表达水平均较正常人显著增高,虽然IL4mRNA及蛋白质的表达与正常人比较无显著性差异,但IL4mRNA/TNFαmRNA和IL4mRNA/IL1αmRNA的比值明显降低,IL4以剂量依赖方式明显抑制LPS诱导的亚重肝患者PBMCsTNFα、IL1的表达;与正常人比较,IL4对亚重肝患者PBMCs的作用效应降低。结论:IL4对亚重肝PBMCsTNFα、IL1的表达均具有明显抑制作用,在亚重肝的治疗上具有潜在的应用价值
Objective: To analyze the regulatory effect of IL4 on TNFα and IL1 in peripheral blood mononuclear cells (PBMCs) in patients with sub-acute severe hepatitis (sub-heavy liver) and to evaluate the potential value of IL4 in the treatment of subhepatic liver.
