目的 :研究人前列腺癌组织中有丝分裂激活蛋白激酶 (MAPK)激活和前列腺癌发生、恶化进展的关系。方法 :采用免疫组织化学分析方法 ,用抗磷酸化MAPK蛋白的抗体检测 4 8例人前列腺癌组织标本中MAPK激活。结果 :所有前列腺癌组织中均有磷酸化MAPK蛋白表达 ,而非癌前列腺组织中未发现有磷酸化MAPK蛋白表达。磷酸化MAPK蛋白表达强阳性在Gleason分值≤ 7和 8~ 10组中分别为 2 0 %和 6 1.1% (P <0 .0 1) ;在局部浸润性肿瘤 (T3 期 )中强阳性率 5 7 1%明显高于局限性肿瘤 (T1~T2 期 ) 2 6 5 % (P <0 .0 1)。结论 :MAPK激活和人前列腺癌的发生及恶化进展密切相关 ,可以作为估价前列腺癌恶性程度和预后的指标。
Objective: To investigate the relationship between the activation of mitogen-activated protein kinase (MAPK) and the progression of prostate cancer in human prostate cancer. Methods: Immunohistochemical analysis was used to detect the activation of MAPK in 48 specimens of human prostate cancer using anti-phospho-MAPK antibody. RESULTS: Phosphorylated MAPK protein was detected in all prostate cancer tissues, whereas no phosphorylated MAPK protein was found in non-cancerous prostate tissues. The strong positive expression of phosphorylated MAPK protein was 20% and 61.1% (P <0.01) in Gleason score≤7 and 8 ~ 10 respectively. The positive rate of phosphorylated MAPK protein in locally invasive tumor (T3) 5 7 1% was significantly higher than that of the localized tumor (T1 ~ T2) by 26.5% (P <0.01). Conclusion: Activation of MAPK is closely related to the occurrence and progression of human prostate cancer, which can be used as an index to evaluate the malignancy and prognosis of prostate cancer.
