从四川自贡某盐井壁植物根系土壤中分离得到1株中度嗜盐解磷菌QW1011。该菌细胞呈线状,大小为0.8μm×30μm~100μm,革兰氏染色为阳性、最适NaCl生长浓度为10%,NaCl最高耐受浓度15%。好氧生长,酪素水解、硝酸还原和接触酶阴性。菌株的16S rRNA基因序列(接受号:EF647207)与Bacillus megatherium ATCC14581的16S rRNA相似性为100%,其16S-23S rRNA间区(ISR)的PCR扩增片的PAGE指纹图谱与参考菌株Bacillus megaterium ATCC14581和Bacillus megatherium var.phosphaticum DSM3228存在较大的差异。基于形态学、生理生化、16S rRNA及ISR的初步分析,认为中度嗜盐解磷菌QW1011为Bacillus属,对其系统分类还需结合其它分类学指标做进一步分析。当温度24°C~36°C,NaCl浓度5%~10%,pH6.5~8.0培养60h时,菌株QW1011对Ca3(PO4)2盐培养基都具有较好的解磷能力,其解磷能力可达56.76μg/mL~71.34μg/mL,且对Ca3(PO4)2盐的解磷能力优于卵磷脂的解磷能力。

The PAGE fingerprint of the 16S rRNA gene region (ISR) of the strain was similar to the reference strain Bacillus megaterium ATCC14581 And Bacillus megatherium var.phosphaticum DSM3228 there is a big difference. Based on the morphological, physiological and biochemical, 16S rRNA and ISR analysis, it is considered that the moderate halophilic phosphorus solubilizing bacteria QW1011 is Bacillus, and its taxonomy needs to be further analyzed in combination with other taxonomic indicators. Strain QW1011 had good ability to dissolve phosphorus in Ca3 (PO4) 2 salt medium when the temperature was 24 ° C ~ 36 ° C, NaCl concentration was 5% ~ 10% and pH was 6.5 ~ 8.0. Its ability is up to 56.76μg / mL ~ 71.34μg / mL, and its ability to dissolve Ca3 (PO4) 2 is better than that of lecithin.
