目的观察内脂素对大鼠肝星状细胞(HSCs)表达碱性成纤维细胞生长因子(FGF-2)的影响,探讨内脂素在肝纤维化中的作用。方法对培养激活的大鼠HSCs分别给予不同浓度(0、50、100和200 ng/mL)的内脂素进行刺激,在不同时限(6 h、12 h、24 h及36 h)收集细胞总RNA。用实时荧光定量(SYBR GreenⅠ)RT-PCR检测FGF-2 mRNA的表达。结果 50 ng/mL内脂素孵育HSCs 24 h可上调FGF-2 mRNA的表达(1.71±0.34 vs.1,P<0.05),随着刺激浓度的增加FGF-2 mRNA的表达进一步上调(P<0.05)。100 ng/mL内脂素孵育HSCs 12h可促进FGF-2 mRNA的表达(1.68±0.31 vs.1,P<0.05),随着刺激时间的延长,FGF-2 mRNA的表达逐渐增加(P<0.05)。结论内脂素能诱导大鼠HSCs表达FGF-2,提示内脂素可能通过诱导FGF-2的表达参与肝纤维化的发病过程。
Objective To investigate the effect of visfatin on the expression of basic fibroblast growth factor (FGF-2) in rat hepatic stellate cells (HSCs) and to explore the role of visfatin in hepatic fibrosis. Methods Rat HSCs cultured in vitro were stimulated with visfatin at different concentrations (0, 50, 100 and 200 ng / mL), and the total cells were harvested at different time points (6 h, 12 h, 24 h and 36 h) RNA. The expression of FGF-2 mRNA was detected by real-time fluorescence quantitative PCR (SYBR GreenⅠ) RT-PCR. Results FGF-2 mRNA expression was up-regulated at 50 ng / mL visfatin in 24 h (1.71 ± 0.34 vs.1, P <0.05). The expression of FGF-2 mRNA was up-regulated with the increase of stimulation concentration (P < 0.05). FGF-2 mRNA expression was enhanced by incubation with 100 ng / mL visfatin for 12 h (1.68 ± 0.31 vs.1, P <0.05), and the expression of FGF-2 mRNA increased gradually with stimulation time ). Conclusion Visfatin can induce the expression of FGF-2 in rat HSCs, suggesting that visfatin may be involved in the pathogenesis of hepatic fibrosis by inducing FGF-2 expression.
