目的探讨Wnt信号通路阻滞剂DKK-1对滋养细胞转录因子GCM-1及Htr A4表达及侵袭力的影响。方法对绒毛膜滋养层细胞系JEG-3细胞施加外源性重组DKK-1,然后分为对照组和DKK-1组(150 ng/ml)。采用实时荧光定量PCR、蛋白印迹技术及Transwell实验分别检测β-链蛋白、GCM-1、Htr A4 mRNA和蛋白的表达情况及细胞侵袭力的变化。结果外源性重组DKK-1干预24 h后,与对照组比较,DKK-1组β-链蛋白、GCM-1和Htr A4 mRNA和蛋白表达水平均明显降低(P<0.05),Transwell实验显示DKK-1组JEG-3细胞的侵袭力明显下降(P<0.05)。结论 Wnt信号通路阻滞剂DKK-1能够减少GCM-1和Htr A4在滋养层细胞中的表达,进而降低滋养细胞的侵袭力。
Objective To investigate the effect of Wnt signaling pathway inhibitor DKK-1 on the expression and invasiveness of gestational trophoblastic transcription factors GCM-1 and Htr A4. Methods Exogenous recombinant DKK-1 was administered to chick trophoblast cell line JEG-3 and then divided into control group and DKK-1 group (150 ng / ml). The expression of β-catenin, GCM-1, Htr A4 mRNA and protein and the invasiveness of cells were detected by real-time fluorescence quantitative PCR, Western blot and Transwell assay respectively. Results Compared with the control group, the mRNA and protein expressions of β-catenin, GCM-1 and Htr A4 in DKK-1 group decreased significantly (P <0.05) 24 h after exogenous recombinant DKK-1 was administered. The invasiveness of JK-3 cells in DKK-1 group was significantly decreased (P <0.05). Conclusion Wnt signaling pathway inhibitor DKK-1 can reduce the expression of GCM-1 and Htr A4 in trophoblast cells, thereby reducing the invasiveness of trophoblast cells.
