【目的】观察醋柳黄酮对结扎家兔冠状动脉前降支所致的心肌缺血再灌注家兔心肌组织中Bcl-2及Bax基因及蛋白表达的影响。【方法】选用24只健康新西兰大白兔随机分为假手术组、模型组、醋柳黄酮组(剂量为0.96 mg.kg-1.d-1)。采用结扎家兔冠状动脉前降支方法复制急性心肌缺血模型,Real-time PCR及Western blot方法检测Bcl-2及Bax基因及蛋白表达变化。【结果】模型组心肌组织Bcl-2、Bax mRNA,Bax蛋白表达显著升高(均P<0.01),Bcl-2蛋白表达显著降低(P<0.01);醋柳黄酮组可显著升高Bcl-2 mRNA及蛋白表达,降低Bax mRNA与蛋白表达,与模型组比较差异有统计学意义(P<0.05)。【结论】醋柳黄酮对家兔冠脉结扎后引起的心肌缺血损伤的保护作用机制可能与其对凋亡基因的调节密切相关。
【OBJECTIVE】 To observe the effects of tiumelitzia flavonoids on the gene and protein expression of Bcl-2 and Bax in myocardium of rabbits with myocardial ischemia-reperfusion injury induced by anterior descending coronary artery ligation in rabbits. 【Methods】 Twenty-four healthy New Zealand white rabbits were randomly divided into sham-operation group, model group and ti-willow-flavone group (dose of 0.96 mg.kg-1.d-1). The model of acute myocardial ischemia was established by ligation of rabbit anterior descending coronary artery. The expression of Bcl-2 and Bax gene and protein were detected by Real-time PCR and Western blot. 【Results】 The expression of Bcl-2, Bax mRNA and Bax protein were significantly increased (all P <0.01) and the protein expression of Bcl-2 in model group was significantly decreased (P <0.01) 2 mRNA and protein expression, decreased Bax mRNA and protein expression, compared with the model group, the difference was statistically significant (P <0.05). 【Conclusion】 The protective mechanism of Trifolvulus flavone against myocardial ischemia-reperfusion injury after coronary artery ligation may be closely related to the regulation of apoptosis gene.
