目的从三邻甲苯磷酸酯(tri-orhto-cresyl phosphate,TOCP)暴露鸡的脊髓组织中筛选有机磷致迟发性神经病(organophosphorus ester induced delayed neuropathy,OPIDN)的相关基因,为进一步探讨OPIDN的发病机制提供目的基因谱。方法罗曼鹤母鸡(Roman hens)随机分为TOCP组、先给苯甲基磺酰氟(phenylmethanesul fonyl fluoride,PMSF)再给TOCP的PMSF前干预组、生理盐水对照组,每组4只。染毒5 d处死实验鸡,低温取脊髓,依次提取总RNA、纯化、转录合成cRNA探针、标记生物素、基因芯片杂交、扫描杂交信号、最后进行基因芯片处理和分析。结果 TOCP组与对照组相比有显著差异,并与PMSF前干预组无显著差异的基因有520个,其中表达差异在3倍以上基因有36个。结论从TOCP暴露鸡脊髓组织中筛选出520个OPIDN相关基因,其中36个基因可能为OPIDN密切相关基因。
Objective To screen the related genes of organophosphorus ester induced delayed neuropathy (OPIDN) from the spinal cord tissues of chickens exposed with tri-orhto-cresyl phosphate (TOCP). To explore the pathogenesis of OPIDN The mechanism provides the gene spectrum of interest. Methods Roman hens were randomly divided into two groups: TOCP group, PMSF pretreatment group and POCF pretreatment group and saline control group, with 4 rats in each group. The experimental chickens were sacrificed 5 d after treatment. The spinal cord was taken at low temperature. The total RNA was extracted sequentially, purified and transcribed to cRNA probe. The biotin and gene chip were hybridized and the hybridization signals were scanned. Finally, gene chip processing and analysis were performed. Results There were 520 genes in TOCP group and control group, and there were 520 genes which were not significantly different from those in PMSF pre-intervention group. There were 36 genes whose expression difference was more than 3 times. Conclusion 520 OPIDN-related genes were screened from TOCP-exposed chicken spinal cord tissues, of which 36 genes may be OPIDN closely related genes.
