目的:用HPLC测定人工北虫草及废料酶解液中核苷和虫草素的含量,判断酶解过程对核苷和虫草素的影响。方法:以AgilentC18色谱柱,甲醇-水为流动相,采用梯度洗脱,流速0.8mL·min-1,检测波长260nm,柱温28℃。结果:样品中尿苷、鸟苷、腺苷和虫草素的加标回收率为92.14%~103.23%,相对标准偏差为0.31%~5.41%。结论:该法简单、快速、准确地测定酶解液中3种核苷和虫草素的含量,人工北虫草废料酶解液中核苷和虫草素含量较高,有进一步开发和利用的价值。
OBJECTIVE: To determine the content of nucleoside and cordycepin in artificial Cordyceps militaris and waste hydrolyzate by HPLC, and to determine the effect of enzymolysis on nucleoside and cordycepin. Methods: Agilent C18 column and methanol-water were used as mobile phases. The gradient elution was performed at a flow rate of 0.8 mL · min-1. The detection wavelength was 260 nm and the column temperature was 28 ℃. Results: The recoveries of uridine, guanosine, adenosine and cordycepin in samples were 92.14% -103.23% with relative standard deviations of 0.31% -5.41%. Conclusion: The method is simple, rapid and accurate for the determination of three kinds of nucleosides and cordycepin in the hydrolyzate. The content of nucleoside and cordycepin in the hydrolyzate of Cordyceps militaris is high, which has the value of further development and utilization.
