目的 探讨组织因子 (TF)表达在原发性结直肠癌侵袭及转移中的作用 ,分析TF对人类大肠癌HT 2 9细胞侵袭能力的影响。方法 应用免疫组织化学染色法研究 85例原发性结直肠癌和 6例结直肠良性腺瘤标本TF表达情况 ,分析TF表达与肿瘤侵袭转移及预后的关系 ;利用构建有正义 /反义TFcDNA的质粒pcDNA3 1/Zeo ,以脂质体法转染HT 2 9细胞 ;转染成功的HT 2 9细胞采用Western印迹分析检测TF表达水平并进行基质胶体外侵袭实验观察细胞侵袭能力的变化。结果85例结直肠癌标本中 4 0例 (4 7 1% )TF表达阳性 ,正常黏膜和结直肠良性腺瘤均为TF阴性表达 ;TF阳性表达与结直肠癌的浸润深度密切相关 (r =0 895 ,P <0 0 1) ;TF阳性表达与结直肠癌的同时性 (r =0 974 ,P <0 0 1)和异时性 (r =0 96 3,P <0 0 1)肝转移均密切相关 ;Logistic回归表明TF表达为结直肠癌肝转移的影响因素 (P <0 0 1) ,多因素回归分析表明TF表达是影响原发性结直肠癌预后的因素之一 (P <0 0 1) ;转染了正义TFcDNA的HT 2 9细胞的TF表达水平较未转染的HT 2 9细胞升高 ,转染了反义TFcDNA的HT 2 9细胞的TF表达水平则降低 ;转染了正义TFcDNA的HT 2 9细胞的侵袭能力较未转染的HT 2 9细胞增强 ,转染了反义TFcDNA的HT 2 9细胞的侵袭能力则减弱。结论TF?
Objective To investigate the role of tissue factor (TF) in the invasion and metastasis of primary colorectal cancer and analyze the effect of TF on the invasiveness of human colorectal cancer HT 2 9 cells. Methods The expression of TF in 85 cases of primary colorectal cancer and 6 cases of colorectal benign adenoma was studied by immunohistochemical staining. The relationship between the expression of TF and tumor invasion, metastasis and prognosis was analyzed. Using the constructed sense / antisense TF cDNA The plasmid pcDNA3 1 / Zeo was transfected into HT 2 9 cells by lipofectamine 2000. The transfected HT 2 9 cells were transfected with HT 2 9 cells. The expression of TF was detected by Western blot analysis and the changes of cell invasion were observed by in vitro matrigel invasion assay. Results Forty four cases (471%) of the 85 colorectal cancer specimens were positive for TF expression, while the normal mucosa and colorectal benign adenoma were negative for TF. The positive expression of TF was closely related to the depth of invasion of colorectal cancer (r = 0 895, P <0.01). The positive correlation between TF expression and colorectal cancer (r = 0 974, P 0 01) and heterotopic (r 0 963, P 0 01) (P <0.01). Logistic regression showed that TF expression was the influencing factor of liver metastasis of colorectal cancer (P <0.01), and multivariate regression analysis showed that TF expression was one of the factors affecting the prognosis of primary colorectal cancer (P < The expression level of TF in HT29 cells transfected with sense TFcDNA was higher than that in untransfected HT2 9 cells and decreased in HT29 cells transfected with antisense TFcDNA. The invasive ability of HT29 cells infected with sense TFcDNA was enhanced compared with that of untransfected HT29 cells, while the invasion ability of HT29 cells transfected with antisense TFcDNA was weakened.
