目的:雌激素对大鼠血浆高同型半胱氨酸(Hcy)水平和心肌雌激素受体(ER)mRNA表达水平的影响。方法:蛋氨酸饮食喂养去势雌性Wistar大鼠构建高Hcy血症动物模型,造模同时给予皮下注射雌激素干预的为预防组,造模后再给予皮下注射雌激素干预的为治疗组。高压液相色谱法测定血浆Hcy水平,逆转录聚合酶链式反应法(RT-PCR)半定量检测心肌ERmRNA表达水平。结果:高蛋氨酸饮食喂养90d后,造模组大鼠Hcy水平明显高于对照组(P<0.05),2组ERα和ERβ基因转录水平差异无统计学意义。预防组血浆Hcy水平低于造模组,心肌ERαmRNA水平高于造模组;预防组和治疗组心肌ERβmRNA水平和造模组比较差异无统计学意义(P>0.05)。结论:预防应用雌激素可以降低血浆Hcy水平,升高心肌ERαmRNA水平,但对其心肌ERβmRNA水平未见影响。
Objective: To study the effect of estrogen on plasma homocysteine (Hcy) and the expression of cardiac estrogen receptor (ER) mRNA in rats. Methods: Methionine diet was fed on female Wistar rats to construct animal models of hyperhomocysteinemia. The model rats were given prophylactic estrogen subcutaneously as model group and estrogen intervention group as control group. Plasma Hcy level was measured by high pressure liquid chromatography. The expression of ERmRNA in myocardium was semi-quantitatively detected by reverse transcription-polymerase chain reaction (RT-PCR). Results: After fed with high methionine diet for 90 days, the level of Hcy in the model rats was significantly higher than that in the control rats (P <0.05). There was no significant difference in the transcriptional level of ERα and ERβ between the two groups. The level of plasma Hcy in prevention group was lower than that in model group, and the level of ERαmRNA in myocardium was higher than that in model group. There was no significant difference in ERβ mRNA level between model group and prevention group (P> 0.05). Conclusion: Prevention of estrogen can decrease plasma Hcy level and increase myocardial ERαmRNA levels, but have no effect on myocardial ERβmRNA level.
