近年来,已有越来越多的药物通过皮肤通道的给药系统,特别是控释系统而应用于临床.美国已有4种药物,即东莨菪碱、硝酸甘油、雌二醇和可乐定被认可,还有一些药物尚在审批中.我国对透皮转运系统的研究亦日益重视.要进一步了解体外研究中透皮系统药物和系统面积、赋形剂以及剂量等不同因素对药物疗效产生的影响,并将这些研究结果应用于临床,必须选择和发展适当的在体透皮吸收动力学模型.鉴此,本文综合各类模型的特点及有关研究以供参考.
In recent years, more and more drugs have been used clinically in the skin channel delivery system, especially the controlled release system. There are four kinds of drugs in the United States, namely scopolamine, nitroglycerin, estradiol and clonidine, There are still some drugs are still in the examination and approval.Our country also pay more and more attention to the research of transdermal delivery system.To further understand the influence of different factors such as drug and system area, excipients and dosage in transdermal system in vitro, And apply these findings to clinical practice, we must select and develop appropriate in vivo kinetics models of transdermal absorption.Accordingly, the characteristics of all kinds of models and related studies are summarized in this paper for reference.
