目的:观察木瓜蛋白酶水溶液对小鼠尾静脉注射急性毒性反应,并测定半数致死量(LD50)。方法:经蛋白酶活力鉴定,本实验所用木瓜蛋白酶水溶液活力为20 158.605U·mg~(-1)。静脉注射木瓜蛋白酶水溶液按组间公比0.931设为6个组,分别为50mg·kg~(-1)、47mg·kg~(-1)、43mg·kg~(-1)、40mg·kg~(-1)、38 mg·kg~(-1)、35 mg·kg~(-1)组,另设对照组,采用Bliss法测定LD50。结果:随剂量升高,木瓜蛋白酶水溶液各给药组小鼠给药后出现自主活动减少,摇头,跳跃,抽搐,挣扎,口鼻流出红色分泌物,直至死亡,小鼠死亡集中于用药后10min内,肺部有大量红色分泌物。雄小鼠LD50为39.49mg·kg~(-1),雌小鼠LD50为40.07mg·kg~(-1),雌雄无明显差异,雌雄合并统计LD50为40.55mg·kg~(-1)。结论:木瓜蛋白酶水溶液小鼠尾静脉注射给药LD50为40.55mg·kg~(-1),95%CI为39.47~41.62mg·kg~(-1),毒性靶器官为肺,雌雄无明显差异。
Objective: To observe the acute toxicity of papain aqueous solution on the tail vein of mice and to determine the LD50. Methods: The protease activity of papain aqueous solution used in this experiment was 20 158.605 U · mg -1. The intravenous injection of papain aqueous solution was divided into six groups according to the male sterility ratio of 0.931, which were 50 mg · kg -1, 47 mg · kg -1, 43 mg · kg -1, 40 mg · kg -1, (-1), 38 mg · kg -1, 35 mg · kg -1, respectively. Another control group was established. The LD50 was determined by Bliss method. Results: With the increase of dose, the mice in each administration group of papain decreased spontaneously, shaking their heads, jumping, twitching and struggling, the red secretions were excreted in the mouth and nose until their death. The death of mice was concentrated in 10 min Inside, the lungs have a lot of red secretions. The LD50 of male mice was 39.49 mg · kg -1, the LD50 of female mice was 40.07 mg · kg -1, there was no significant difference between male and female, LD50 was 40.55 mg · kg -1. Conclusion: The LD50 of papain aqueous solution is 40.55 mg · kg ~ (-1), and the 95% CI is 39.47 ~ 41.62 mg · kg ~ (-1). The toxic target organ is lung, there is no significant difference between male and female .
