目的建立视网膜母细胞瘤细胞株SO-RB50皮下移植的裸鼠动物模型,观察白藜芦醇对肿瘤的影响。方法以2×108/ml SO-RB50细胞皮下注射制备视网膜母细胞瘤皮下移植动物模型。造模成功后将裸鼠随机分为实验组和对照组,实验组给予白藜芦醇、对照组给予等量生理盐水。测量裸鼠肿瘤体积并比较两组肿瘤重量;MTT法检测肿瘤细胞活性;蛋白印迹(Western blot)技术检测肿瘤增殖细胞核抗原(PCNA)、基质金属蛋白酶-2(MMP-2)、MMP-9的表达。结果与对照组比较,实验组肿瘤生长明显减慢,体积小于对照组(t=2.283,P<0.05);实验组肿瘤重量低于对照组,差异有统计学意义(t=4.535,P<0.05)。MTT结果显示实验组肿瘤细胞活性低于对照组,差异有统计学意义(t=8.764,P<0.05);Western blot结果显示,实验组PCNA、MMP-2、MMP-9蛋白表达均明显低于对照组,差异有统计学意义(t=5.812、7.077、6.216,P<0.05)。结论白藜芦醇可有效抑制SO-RB50细胞裸鼠皮下移植瘤的生长。
Objective To establish a nude mouse model of retinoblastoma cell line SO-RB50 transplanted subcutaneously to observe the effect of resveratrol on the tumor. Methods Subcutaneous injection of 2 × 108 / ml SO-RB50 cells into human retinoblastoma cell model was established. After successful modeling, nude mice were randomly divided into experimental group and control group, experimental group given resveratrol, and control group were given the same amount of saline. The tumor volume was measured and the tumor weights were compared between the two groups. The activity of tumor cells was detected by MTT assay. The expressions of proliferating cell nuclear antigen (PCNA), matrix metalloproteinase-2 (MMP-2), MMP-9 expression. Results Compared with the control group, the tumor growth in the experimental group was significantly slower than that in the control group (t = 2.283, P <0.05). The tumor weight in the experimental group was lower than that in the control group (t = 4.535, P <0.05) ). The results of MTT showed that the activity of tumor cells in the experimental group was lower than that in the control group (t = 8.764, P <0.05). Western blot results showed that the expression of PCNA, MMP-2 and MMP-9 in the experimental group was significantly lower than that in the control group Control group, the difference was statistically significant (t = 5.812,7.077,6.216, P <0.05). Conclusion Resveratrol can effectively inhibit the growth of subcutaneous xenografts in SO-RB50 cells.
