目的:优选D101大孔树脂纯化红花中羟基红花黄色素A的工业化放大工艺。方法:采用HPLC测定羟基红花黄色素A含量,通过单因素试验考察上样液pH、洗脱剂体积分数和用量、药材-树脂质量比对纯化工艺的影响,通过中试及工业化生产验证优选的纯化工艺。结果:优选的纯化工艺条件为上样液pH 4.0,药材-树脂质量比1∶6,加4 BV水以3 BV.h-1的流速洗脱除杂,用5%乙醇6 BV以3 BV.h-1的流速洗脱,收集洗脱液。结论:该工艺简单可行、分离效果好,适用于羟基红花黄色素A的大生产。
OBJECTIVE: To optimize the industrial scale-up process of hydroxysafflor yellow A in D101 macroporous resin purified safflower. Methods: The content of hydroxysafflor yellow A was determined by HPLC. The effects of pH value, volume fraction and dosage of eluent on the purification process were investigated by single factor test. Purification process. Results: The optimal purification conditions were as follows: the sample solution pH 4.0, the ratio of drug to resin 1: 6, 4 BV water at 3 BV.h-1, eluting at a flow rate of h-1 RTI> and collecting the eluate. Conclusion: The process is simple and feasible, the separation effect is good, suitable for large-scale production of hydroxysafflor yellow A
