人参皂苷Rb1对胎鼠背根神经节P物质表达变化的影响

目的观察不同浓度人参皂苷Rb1对胎鼠背根神经节(DRG)神经元生长状态及P物质(SP)表达的影响。方法选择胎龄为15天的SD大鼠,获取DRG神经元并进行体外分散培养,48 h后,随机分为对照组、低浓度Rb1(10μg/ml)组、中浓度Rb1(100μg/ml)组、高浓度Rb1(1 000μg/ml)组,继续培养12 h,倒置相差显微镜观察各组神经元的生长状态,免疫组织化学方法检测SP在各组中的表达。结果对照组DRG神经元细胞贴壁呈单层散在分布,突起互相交织形成网状。不同Rb1添加组DRG神经元细胞部分呈簇状聚集,突起较长且相互交织。免疫组化结果显示SP主要表达于DRG细胞质中,添加Rb1后,SP的表达明显多于正常对照组,比较有统计学意义(P<0.05)。随着Rb1浓度的增加,SP表达量没有明显差异。结论人参皂苷Rb1可以促进胎鼠DRG神经元的生长,且能够调节DRG神经元SP的表达,对胎鼠背根神经节神经元具有一定的保护作用。 Objective To observe the effects of different concentrations of ginsenoside Rb1 on the growth of neurons and the expression of substance P in fetal rat dorsal root ganglion (DRG) neurons. Methods SD rats with gestational age of 15 days were selected, and DRG neurons were obtained and cultured in vitro. After 48 hours, they were randomly divided into control group, low concentration of Rb1 (10μg / ml), medium concentration of Rb1 (100μg / ml) Group and Rb1 (1 000μg / ml) group. The growth of neurons in each group was observed by inverted phase contrast microscope. The expression of SP in each group was detected by immunohistochemical method. Results The DRG neurons in the control group showed monolayer scattered distribution, and the protrusions interweaved to form a network. Different Rb1 addition group DRG neuron cells were clustered clusters, protrusions longer and intertwined. The results of immunohistochemistry showed that SP was mainly expressed in the cytoplasm of DRG. After addition of Rb1, the expression of SP was significantly more than that of normal control group (P <0.05). With the increase of Rb1 concentration, SP expression did not differ significantly. Conclusion Ginsenoside Rb1 can promote the growth of fetal rat DRG neurons and regulate the expression of SP in DRG neurons, which may protect neurons in the dorsal root ganglion of fetal rats.
