目的研究莫诺苷对缺血再灌注7 d大鼠皮层中金属基质蛋白酶组织抑制剂(tissue inhibitor of metalloproteinase,TIMP)表达的影响。方法 25只雄性SD大鼠采用改良Zealonga线栓法制备大鼠大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)模型,术后随机分为模型组、莫诺苷小剂量组、中剂量组、大剂量组,每组5只。造模3 h后按30、90、270 mg/kg剂量每天一次灌胃莫诺苷。免疫印迹法(Western Blotting)和免疫荧光染色法(Immunofluorencent staining)分析莫诺苷对脑缺血再灌注7 d大鼠患侧皮层TIMP表达的影响。结果脑缺血再灌注7 d后,与假手术组相比,模型组TIMP蛋白表达显著升高(P<0.01);同模型组相比,莫诺苷中剂量组(90 mg/kg)和大剂量组(270 mg/kg)TIMP蛋白表达均明显升高(P<0.05;P<0.01)。结论莫诺苷可以上调TIMP在大脑皮层内的表达,有助于脑卒中后血脑屏障的保护及修复。
Objective To investigate the effects of morroniside on the expression of tissue inhibitor of metalloproteinase (TIMP) in the cortex of rats 7 d after ischemia-reperfusion. Methods Twenty-five male Sprague-Dawley rats were used to establish the middle cerebral artery occlusion (MCAO) model by modified Zealonga thread occlusion method. The rats were randomly divided into model group, low dose morroniside group, middle dose group, Dose group, each group of five. After 3 h of modeling, morroniside was intragastrically administered at doses of 30, 90 and 270 mg / kg. The effect of morroniside on the expression of TIMP in ipsilateral cortex of cerebral ischemia-reperfusion 7 d rats was analyzed by Western Blotting and Immunofluorescence staining. Results Compared with the sham operation group, the TIMP protein expression in model group was significantly increased (P <0.01) on the 7th day after cerebral ischemia / reperfusion. Compared with the model group, the median dose of morroniside (90 mg / kg) and TIMP protein expression in high dose group (270 mg / kg) was significantly increased (P <0.05; P <0.01). Conclusion Morroniside can up-regulate the expression of TIMP in the cerebral cortex and contribute to the protection and repair of the blood-brain barrier after stroke.
