目的:建立反相高效液相色谱法同时测定莪术油中莪术二酮、吉马酮、呋喃二烯3种倍半萜类成分含量。方法:采用Agilent Extend C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-水(B),梯度洗脱(0~20 min,60%A→95%A;20~35 min,95%A),流速1.0 mL.min-1;检测波长216 nm;柱温30℃。结果:莪术二酮、吉马酮和呋喃二烯浓度分别在68.04~3402 ng(r=1.000),26.02~1301 ng(r=1.000),40.14~2007 ng(r=1.000)范围呈现良好的线性关系;平均回收率(n=6)分别为102.0%(RSD=0.69%),101.2%(RSD=0.65%),99.70%(RSD=0.54%);重复性试验,3种成分含量的RSD(n=6)均小于1.0%。结论:本方法快速准确,具有良好的重复性和稳定性,可用于莪术油的质量控制。
Objective: To establish a reversed-phase high performance liquid chromatographic method for the simultaneous determination of three sesquiterpenoids in Curcuma dione, gemmazone, and furandi in Ezhu oil. Methods: Agilent Extend C18 column (4.6 mm×250 mm, 5 μm) was used. The mobile phase was acetonitrile (A)-water (B). Gradient elution (0~20 min, 60% A→95% A; 20~ 35 min, 95% A), flow rate 1.0 mL.min-1; detection wavelength 216 nm; column temperature 30°C. RESULTS: The concentration of curcumone dione, geimagone, and furin was 68.04 to 3402 ng (r=1.000), 26.02 to 1301 ng (r=1.000), and 40.14 to 2007 ng (r=1.000). Relationships; The average recovery (n=6) was 102.0% (RSD=0.69%), 101.2% (RSD=0.65%), and 99.70% (RSD=0.54%); Reproducibility test, RSD of 3 component contents (respectively). N=6) are less than 1.0%. Conclusion: The method is rapid and accurate, with good repeatability and stability, and can be used for quality control of zedoary oil.
