目的:比较两株肺癌细胞系(经实验证明为高、低侵袭系)体外培养细胞和裸鼠体内移植瘤细胞的增殖状态;方法:采用激光流式细胞仪,测量单个完整细胞群体的DNA含量,分析其各细胞周期的比率;结果:体外培养的高增殖系细胞,移植入裸鼠体内后其G2/M期细胞比率反而降低,体外培养的低增殖系细胞,移植入裸鼠体内后其G2/M细胞比率反而升高,两株肺癌细胞系的增殖指数间无显著差异;结论:两株肺癌细胞系在体内、外增殖状况的差异性,说明肿瘤细胞在体外增殖速度的快慢不能准确反映其在体内的增殖状态,必须结合肿瘤细胞的其它生物学特性,才能比较准确地分析和判断肿瘤细胞的恶性程度。
OBJECTIVE: To compare the proliferation of two lung cancer cell lines (high and low invasion lines as experimentally demonstrated) in vitro and in vivo in nude mice: Methods: Laser flow cytometry was used to measure the DNA content of a single intact cell population. , Analysis of the ratio of each cell cycle; Results: In vitro cultured highly proliferative cells, transplanted into nude mice, the G2/M phase of the cell rate instead decreased, in vitro cultured low proliferative cells transplanted into nude mice The ratio of G2/M cells increased but there was no significant difference in the proliferation index between the two lung cancer cell lines. Conclusion: The difference in the proliferation of two lung cancer cell lines in vitro and in vivo indicates that the proliferation rate of tumor cells in vitro is not accurate. To reflect its proliferation status in the body, it must combine with other biological characteristics of tumor cells to accurately analyze and judge the degree of malignancy of tumor cells.
