为探讨含dUTP/尿嘧啶DNA糖基化酶(dUTP/UDG)PCR试剂(抗污染PCR试剂,PCR-dUTP/UDG)抗PCR产物污染的能力及其在临床检测中的应用,采用该试剂检测204份病毒性肝炎患者血清中HBVDNA,并与普通PCR试剂比较。结果显示:抗污染PCR试剂有较强的抗污染能力,至少可阻止100ng的PCR产物的污染。该试剂与地戈辛素探针分子杂交所测结果的符合率(89.32%)高于普通PCR与分子杂交所测的符合率(81.45%)。表明抗污染PCR试剂的应用有利于防止PCR产物的污染,提高PCR的准确性和特异性。
To investigate the ability of dUTP / UDG PCR reagents (anti-contamination PCR reagents, PCR-dUTP / UDG) to resist the contamination of PCR products and their application in clinical detection, this reagent was used to detect 204 serum HBV DNA in patients with viral hepatitis, and compared with ordinary PCR reagents. The results showed that the anti-contamination PCR reagent has strong anti-pollution ability, at least can prevent the contamination of 100ng PCR product. The coincidence rate (89.32%) between the reagent and Digoxigenin probe hybridization results was higher than that of the normal PCR and molecular hybridization (81.45%). It shows that the application of anti-contamination PCR reagent is conducive to prevent the contamination of PCR products and improve the accuracy and specificity of PCR.
